Cloning and Expression of Human Gamma-Interferon cDNA in E. coli
Authors: not saved
Abstract:
Prior to the production of human gamma interferon using recombinant DNA technology, it had been producedmainly upon mitogenic induction of lymphocytes in very low amounts, which evidently hamperedits characterization and its medical applications. The recombinant gamma interferons produced in largerquantities in prokaryotic systems retain their biological activities, and can be used clinically in the treatmentof various viral, neoplastic and immunosuppressed conditions or diseases. In this study, a cDNAsequence coding for human gamma interferon was synthesized from mRNA template extracted frominduced human T lymphocytes. The cDNA was then amplified by PCR, cloned in an expression vector, and transformed into Escherichia coli. The polypeptide produced through the expression of this DNAsequence in E. coli showed immunological and chemical properties resembling authentic human IFN-γ.

The plasmid pALCA1SIFN containing cDNA that encodes the human interferon a-2b was obtained from the ATCC(no. 531667). In this system the expression of the gene is under the control of an alcA promoter. alcA p is a specific promoter for expression of different genes in Aspergillusfilamentous. In this plasmid the coding region of IFN?-2b is preceded by the coding region of a synthetic signal ...

Background: IFN-g is mostly secreted by activated CD4+ , CD8+ T cells and NK cells. This cytokine has immunomodulatory, anti-cancer and anti-microbial effects and is important for prophylaxis, diagnosis and treatment of chronic infections and cancers. Objective: The purpose of this study was to clone the full cDNA of human IFN-g and express it in CHO cell line. Methods: Lymphocytes from a healt...
full textInducible Expression of Human Gamma Interferon
Background:The premature termination of high producer clones, which will be killed due to cell proliferation and proteins production antagonism, is one of the basic drawback in recombinant proteins technology. Furtheremore, it is supposed some toxic proteins like interferon which we intended to clone and express, inhibit host cells’ proliferation. So, it is necessary to tightly control IFN-γ pr...

Background: Leukemia inhibitor factor (LIF) is a very important pleiotropic cytokine which belongs to interleukin-6 (IL-6) family. LIF exerts multiple effects on different types of cells and tissues with numerous regulatory effects in vivo and in vitro. It is a lymphoid factor, which performs a number of activities including cholinergic neuron differentiation, contro...
